Description
Ipamorelin Overview
Ipamorelin is a pentapeptide consisting of Aib-His-D-2-Nal-D-Phe-Lys-NH2. This Peptide effectively stimulates the pituitary gland to release growth hormone and supports lean muscle mass growth and weight loss. Additionally, it has minimal impact on:
- thyroid-stimulating hormone
- luteinizing hormone
- prolactin
- follicle-stimulating hormone
- ACTH
- cortisol release
Ipamorelin is a member of the Growth Hormone-Releasing Peptides (GHRP) category, recognized for their capacity to boost growth hormone levels and provide numerous clinical benefits, including:
- The management of body fat and weight
- The enhancement of skin elasticity and function
- The reduction in cortisol production
- The enhancements in energy levels and sleep quality
Ipamorelin is often correlate to GHRP-6 and GHRP-2 due to their similar mechanisms, actions, and functionalities.
What Is Ipamorelin?
Ipamorelin is a Peptide made up of five amino acids, primarily recognized as a secretagogue, meaning it stimulates the production of specific hormones. It promotes the secretion of growth hormone from the pituitary gland and mimics ghrelin, the hunger hormone produced in the stomach.
Ipamorelin Induces Longitudinal Bone Growth In Rats
Ipamorelin, a potent synthetic pentapeptide, possesses specific growth hormone (GH)-releasing properties. In a study on adult female rats, ipamorelin was injected subcutaneously at varying doses three times daily for 15 days to investigate its effects on longitudinal bone growth rate (LGR), body weight (BW), and GH release.
Results showed that ipamorelin dose-dependently increased longitudinal bone growth rate (LGR), with rates rising significantly compared to the control group. There was also a significant and dose-dependent effect on body weight (BW) gain. However, ipamorelin treatment did not influence total IGF-I levels, IGFBPs, or serum markers of bone formation and resorption. The number of tartrate-resistant acid phosphatase-positive multinuclear cells in the tibia’s metaphysis did not change significantly with treatment. [4]
The responsiveness of the pituitary to a provocative intravenous dose of ipamorelin or GHRH showed a marginal reduction in plasma GH response after ipamorelin, but no change after GHRH. The pituitary GH content remained unaffected by ipamorelin treatment. [4]
Comparative Study Of Ipamorelin And GHRP 6 On Bone Mineral Content In Female Rats
The study investigated the effects of growth hormone (GH) secretagogues (GHSs) on bone mineral content (BMC) in young adult female rats. Rats were given ipamorelin (IPA), GH-releasing Peptide-6 (GHRP-6), GH, or vehicle continuously via osmotic minipumps for 12 weeks. Dual X-ray absorptiometry (DXA) measurements were taken every 4 weeks, and femurs were analyzed by mid-diaphyseal peripheral quantitative computed tomography (pQCT) scans. Femurs and vertebrae L6 were analyzed using Archimedes’ principle and ash weight determinations.
All treatments resulted in higher body weight and increased total tibial and vertebral BMC as assessed by DXA compared to the control group. However, total BMC adjusted for body weight increase showed no significant change. Tibial area bone mineral density (BMD) saw an increase, while total and vertebral area BMDs showed no alteration. pQCT measurements indicated that the rise in cortical BMC was due to an enlarged cross-sectional bone area, with cortical volumetric BMD unchanged. Femur and vertebra L6 volumes expanded, with no impact on volumetric BMDs. Ash weight rose with all treatments, while mineral concentration remained steady. [5]
In conclusion, administration of the GHSs ipamorelin and GHRP-6 boosted BMC measured via DXA in vivo. In vitro evaluations using pQCT and Archimedes’ principle, combined with ash weight assessments, revealed that enhancements in cortical and total BMC were attributed to enlarged bone dimensions, while volumetric BMD remained unaltered.
Efficacy Of Ipamorelin In Rodent Model Of Postoperative Ileus
The research examined how ipamorelin, a specific growth hormone secretagogue and ghrelin receptor agonist, might enhance gastrointestinal transit and alleviate symptoms in a rodent model of postoperative ileus (POI). Male rats underwent laparotomy and intestinal manipulation, followed by fasting and infusion of a dye marker into the proximal colon to measure postsurgical colonic transit time.
Ipamorelin, in combination with growth hormone-releasing Peptide (GHRP)-6 or saline (vehicle), was administered via intravenous bolus infusion following either a single dose or a repetitive 2-day dosing regimen. The study assessed outcomes by monitoring the time to the initial bowel movement, cumulative fecal output, food intake, and body weight changes throughout a 48-hour post-surgery period. [6]
The findings revealed that administering a single dose of ipamorelin or GHRP-6 reduced the time to the initial bowel movement compared to the control group. However, there were no significant impacts observed on cumulative fecal output, food intake, or body weight gain over the 48-hour period. [6]
In contrast, repeated administration of ipamorelin notably enhanced cumulative fecal pellet output, food intake, and body weight gain. These results indicate that multiple intravenous infusions of ipamorelin could potentially alleviate symptoms associated with postoperative ileus.
Side Effects from Glucocorticoids
Ipamorelin demonstrates promise in mitigating or reversing the adverse effects associated with glucocorticoid use. Prolonged glucocorticoid administration can lead to inflammatory disorders like cancer and autoimmune diseases, along with reductions in lean muscle mass and intramuscular amino acids, coupled with increased body fat. Ipamorelin’s stimulation of the pituitary gland may potentially counteract these effects.
Muscle Wasting
Glucocorticoids can induce muscle wasting as a side effect. Recent rat studies administering glucocorticoid treatment showed a reduction in amino acid wastage in the liver, alongside improvements in nitrogen balance levels within muscles.
Bone Loss Protection
Prolonged use of glucocorticoids can lead to reduced bone density, increasing the risk of fractures. Clinical studies have highlighted Ipamorelin’s potential to mitigate corticosteroid-induced bone loss through its impact on the pituitary gland. This beneficial effect was observed in rat experiments, where the Peptide actively promoted bone formation. Moreover, Ipamorelin has shown promise in enhancing bone mineral density by strengthening both existing and newly formed bone structures.
Potential Benefits for Diabetes
Studies on diabetic rats indicate that ipamorelin administration may enhance insulin release by binding to pancreatic cell calcium channels. These effects on the pancreas hold promise for potential preventive strategies against type 2 diabetes.
Diagnostic Applications in Cancer and Heart Failure
Ipamorelin, a Peptide known for its interaction with the ghrelin receptor, exhibits heightened affinity in specific contexts, notably in certain cancer types and instances of heart failure. Current research aims to explore Ipamorelin’s potential as a positron emission tomography (PET) probe to enhance diagnostic capabilities for these conditions. Initial in vitro studies have provided promising evidence supporting Ipamorelin’s efficacy as a PET probe.
