Kisspeptin is a hormone-signaling protein naturally produced in humans. In the brain, kisspeptin signaling controls hormone secretion related to the reproductive system.
Scientists are actively studying how kisspeptin impacts testosterone levels and sex-related behaviors, including sexual drive and motivation.
Kisspeptin 10 and Regulation of Testosterone Levels
Normalization of Sex Hormones
Certain forms of kisspeptin have been found to regulate the release of LH in men, helping to normalize sex hormone levels. Administration of kisspeptin-10 to healthy men has resulted in increased LH and testosterone levels. However, at high doses, kisspeptin-10 can cause disruption, leading to continuous LH release.
Kisspeptin 10 and Energy Balance and Reproduction
Kisspeptin production and release influence energy levels and may regulate energy balance. Studies on mice lacking the kisspeptin receptor (Kiss1r) have shown increased fat tissue and reduced energy levels.
This is because kisspeptin receptors are found in both white and brown fat tissue, suggesting a connection between kisspeptin and energy balance. The interplay between energy levels and reproductive health indicates that kisspeptin plays a role in energy modulation during reproduction.
Anti-Cancer Properties of Kisspeptin-10
Suppression of Melanoma Spread
Kisspeptin can suppress the spread of melanoma, a malignant form of skin cancer. It achieves this by reducing the cancer cells’ ability to bond and preventing them from invading other tissues.
This aids in reducing cancer cell migration. Lower levels of kisspeptin have been found in various metastatic cancer types, including breast, bladder, gastrointestinal, prostate, pancreatic, ovarian, skin, and thyroid cancers.
Connection with Melatonin and Cancer Prevention
An experiment in 2020 revealed a connection between kisspeptin, melatonin, and cancer prevention. Mice exposed to daylight showed high kisspeptin levels and low melatonin levels, compared to mice kept in the dark.
Despite the daylight-exposed mice exhibiting high tumor growth rates and volume, no metastasis was observed. This study supports the potential of melatonin and kisspeptin in suppressing tumor spread.
Kisspeptin 10 Cognitive Effects and Behavior
Kisspeptin influences areas of the brain responsible for memory consolidation and 3-D space orientation. Administering kisspeptin Peptides to mice has been shown to reverse learning and navigational impairments associated with alcohol intoxication.
Additionally, studies have demonstrated that kisspeptin administration to healthy heterosexual men enhances limbic brain activity, leading to increased reward-seeking behavior, drive, and overall mood improvement.
Role of Kisspeptin-10 in Kidney Health and Development
Kisspeptin and its receptor are present in various locations throughout the kidney, playing a crucial role in kidney health and development by influencing kidney function signaling. In mice, kisspeptin has been shown to enhance the growth of nerves and blood vessels within the kidney.
Furthermore, kisspeptin may regulate blood vessel function in specific areas of the body, such as the heart, and potentially help reduce tumor metastasis.
Interactions Between Kisspeptins & Neurokinin B
Kisspeptin and neurokinin B (NKB), encoded by the Kiss1 and TAC3/Tac2 genes, play key roles in regulating crucial reproductive milestones, such as the onset of puberty. Dysfunctional genes or kisspeptin receptors associated with these neuropeptides can lead to sexual maturation issues and infertility in both humans and rodents.
These neuropeptides, along with dynorphin A, are co-expressed in the arcuate nucleus as KNDy neurons. NKB’s stimulatory effects on LH release, mediated by auto-synaptic inputs, induce gonadotropin-releasing hormone (GnRH) secretion in a kisspeptin-dependent manner.
Influence of Kisspeptin 10 on Milk Synthesis in Bovine Mammary Epithelial Cells
The study aimed to uncover the mechanism by which Kisspeptin-10 (Kp-10), a Peptide hormone, influences milk synthesis in bovine mammary epithelial cells (BMECs), as its role in this process remains unclear. Researchers utilized various approaches, including a GPR54 inhibitor, SIRT6 overexpression plasmid, and siRNA, to investigate the regulatory mechanisms of Kp-10 in milk protein and milk fat synthesis.
The findings revealed that treatment with 100 nM Kp-10 significantly increased milk synthesis in BMECs. Notably, SIRT6 overexpression substantially reduced milk protein and milk fat synthesis in these cells. Additionally, SIRT6 overexpression reversed the activation of the Kp-10-induced mTOR signaling pathway, which is crucial for protein synthesis and cellular growth.
Further analysis suggested that SIRT6 might exert its regulatory effect on the mTOR signaling pathway at the transcriptional level. The results strongly indicate that the Kp-10/GPR54 axis activates the mTOR signaling pathway by suppressing SIRT6 expression, thereby promoting milk synthesis in BMECs.
This comprehensive understanding of the molecular mechanisms underlying Kp-10’s regulation of milk synthesis sheds light on potential targets for enhancing milk production and quality in dairy animals.
Kisspeptin Administration on Adiponectin, Leptin, and Resistin Secretion in Adult Male Rhesus Monkey
In recent years, kisspeptin-KISS1R signaling has emerged as a key regulator of reproductive function across various vertebrate species. Although traditionally associated with reproductive tissues, both KISS1 (which encodes kisspeptin) and its receptor, KISS1R, are expressed in multiple tissues, including adipose tissue. However, the specific role of kisspeptin in adipocyte function remains unclear.
In this study, the effects of exogenous human Kisspeptin-10 on three important adipokines – adiponectin, leptin, and resistin – were investigated in intact adult male rhesus monkeys under different feeding conditions: normal fed, 24-hour fasting, and 48-hour fasting. Fasted monkeys exhibited decreased plasma resistin and leptin levels (p<0.01), while adiponectin levels increased (p<0.05).
Kisspeptin administration significantly increased mean plasma adiponectin levels (p<0.05) under fed and 24-hour fasting conditions compared to pretreatment or vehicle-treated levels. A stimulatory effect was also observed on plasma adiponectin levels under 48-hour fasting conditions, although it was not statistically significant. No significant effect of kisspeptin was observed on mean plasma leptin and resistin levels.
These findings reveal a novel role for kisspeptin as a regulator of adipocyte function in higher primates, specifically by stimulating plasma adiponectin levels under fed and 24-hour fasting conditions in adult male rhesus monkeys.

Kisspeptin Peptide FAQs
What Does Kisspeptin-10 Do?
Kisspeptin 10 stimulates gonadotropin release in males and in females during the preovulatory phase of the menstrual cycle, but not during the follicular phase.
How Much Does Kisspeptin-10 Increase Testosterone?
Kisspeptin 10 infusion at 4 μg/kg/hr for 22.5 hours increased serum testosterone levels by 50% in both female and male test subjects.
Is Kisspeptin 10 a Natural Indicator of Pregnancy?
Clinical data indicates that Kisspeptin 10 measurement may be more accurate than hCG in differentiating between miscarriage and viable pregnancy after 6 weeks of gestation.
What Is the Role of Kisspeptin 10 in the Activation of Gonadotropin-Releasing Hormone GnRH Neurons?
Kisspeptin 10 is a potent stimulator of GnRH neurons, acting through the G protein-coupled receptor GPR54. Kisspeptin signaling increases the excitability of GnRH neurons and promotes the release of GnRH into the portal circulation.
GnRH then travels to the anterior pituitary gland, stimulating the secretion of the gonadotropins luteinizing hormone (LH) and follicle-stimulating hormone (FSH). These hormones are essential for the development and function of the gonads and the production of sex hormones.
How Does Kisspeptin 10 Signaling Affect the Development and Progression of Central Precocious Puberty (CPP)?
Kisspeptin 10 signaling is believed to play a role in the development and progression of central precocious puberty (CPP) by activating GnRH neurons and stimulating the release of gonadotropins. This activation can result in early gonadal development and the onset of puberty at a young age.
Central precocious puberty (CPP) can negatively impact a child’s physical and emotional development. Therefore, understanding the underlying mechanisms of the condition and how it can be treated is crucial.
